Regulatory T Cell Research
Focus Inhibitory Cytokines

Mouse and Human IL-35

  • Predominantly Secreted by Regulatory T Cells
  • Heterodimer Composed of the p35 Subunit of IL-12 and EBI3
  • Induces Regulatory T Cell (Treg) Proliferation
  • Required for Maximum Suppressor Function of Mouse & Human Tregs
  • Suppresses T Cell Proliferation, Function & Differentiation to Effector
    T Cell Lineages
  • Induces the Conversion of Naïve T Cells into iTr35 Cells (Treg induced by IL-35)
  • Limits Anti-Tumor Immunity
  • Suppresses Autoimmune and Inflammatory Responses
  • Potential Diagnostic Biomarker for Cancers and Chronic Inflammation

PRODUCT SPECIFIC LITERATURE REFERENCES:

• Interleukin-35 administration counteracts established murine type 1 diabetes - possible involvement of regulatory T cells: K. Singh, et al.; Sci. Rep. 5, ID12633 (2015) [Reports the use of AdipoGen's IL-35 (mouse):Fc (human) (CHI-MF-11135) to reverse established hyperglycemia in a NOD mouse model of Type 1 Diabetes (T1D)].

• Cytokine modulation by IL-35 in mice with allergic rhinitis: M. Yokota, et al.; Am. J. Rhinol. Allergy 29, 251 (2015) [Used AdipoGen's IL-35 (mouse):Fc (human) (CHI-MF-11135) to inhibit allergic T cell response in a mouse model of allergic rhinitis].

Figure: Effect of IL-35 (mouse):Fc (human) (rec.) (Prod. No. CHI-MF-11135) on IL-2 expression. Method: Mice were injected with OVA and alum. Splenic CD4+CD25- T cells and DCs were isolated and cultured in the presence of OVA with IL-35 (mouse):Fc (human) (rec.) (10ng/ml) or without IL-35 (mouse):Fc (human) (rec.) (control). IL-2 levels in the supernatant were measured using an IL-2 specific ELISA.

Unique - Highly Active TGF-β1 Mutant

TGFβ1 (mutant) (human):Fc (human) (rec.) CHI-HF-210TGFBM

This product is a long-lasting biologically active form of TGF-β1, where three cysteine codons in the proregion of human TGF-β1 precursor have been changed into serine (Ser) codons. The mutant TGF-β1 is fused to human Fc to produce an auto-active TGF-β1/Fc immunoligand that does not depend on acidification for activation. TGF-β1:Fc mutant has a long half-life in vivo (>32h versus 3min for non-Fc version of TGF-β1).

LIT: Combined administration of a mutant TGF-beta1/Fc and rapamycin promotes induction of regulatory T cells and islet allograft tolerance: W. Zhang, et al.; J. Immunol. 185, 4750 (2010)
